Method

  1. Count each seed quantity and label petri dishes using clear tape and sharpie. 15/20(TBD) seeds per petri dish. 4 repetitions for each variety. Total of 20 petri dishes.
  2. Place equal sized filter paper pieces in the bottom of each petri dish. Each filter paper should cover the bottom surface area of each dish.
  3. Dampen each filter paper by pouring 1 Tablespoon of deionized H20 in each petri dish. Swirl each dish so water is evenly coated and let sit to absorb for a few seconds.
  4. Evenly place each seed group(s) in trays, making sure there is no overlapping.
  5. Moisten specimens with 1 T DI water per petri dish & record data every other day.
    1. Record code data for each sample every other day. Code description will be marked per seed and as percentages % for each seed sample.
    2. When radicle is present, measure radicle length in mm – from the initial protrusion on the seed to the tip of the root.

Germination Chamber Data Collection

  • Code for each petri dish sample. A code will be collected based on results pertaining to 50% of each petri sample.
  • Emergence count for each petri sample.
    • Code 07: Hypocotyl with cotyledons or shoot breaking through seed coat.
  • If present Radicle length (mm) of each petri sample.
